The guile finger spinner turned the game for Greater Northern Raiders, picking up a clinical 4-6 off four overs at the Kingston Twin Ovals.
Kingborough lost its final seven wickets for five runs to be dismissed for a paltry 45.
While skipper Sam Cotton removed both openers after their sound 25-run stand, the 19-year-old wreaked havoc with 3-0 off her final 11 balls.
The 19-year-old had No.3 Nikki Carter stumped before deceiving the next three victims with flight clean bowled.
"She was given an opportunity with the ball and took it which was magnificent to see," co-coach Darren Simmonds said.
"She just turned that game on its head really."
The Knights' tail looked stunned, the next three batters run out including off Matthews's second-last ball.
The Raiders claimed an easy nine-wicket victory at a run-a-ball when Matthews's South Launceston teammate Caitlyn Webster hit her fifth boundary for 28 off 23 balls.
Matthews played no role in the Raiders back-to-back win amid a rain-affected clash.
After a delayed start that reduced the T20 to 19 overs, players were sent off 46 balls into University's run chase and on their return the ball was too damp for the spinner.
The Raiders had made 5-122 after an 82-run partnership for the second wicket.
Ulverstone star Montana Bradley fell for 39, sparking three wickets to fall in the space of seven deliveries.
Riverside's Ianthe Boden was run out for 49 looking for an extra run off the last ball.
Uni were 3-34 when rain came down after ex-Launceston star Katelyn Fryett was run out for 18, requiring 49 more off 4.2 overs under the Duckworth-Lewis method.
The Lions were kept to 7-62 in a 20-run loss and a third win for the Raiders from four.
"We're very happy to come down on our first road trip," Simmonds said.
"We were looking to play really good cricket and bounce back after losing our last game."
